Looks at surface expression of T cell receptor V beta families. In normal individuals at least 70 % of their T cells will be detected with the panel of antibodies used, and there will be a polyclonal distribution. Oligoclonal expansions on a polyclonal background are suggestive of infection or malignancy, whilst an oligoclonal distribution is seen in defects of T cell development
